There are two synthesis methods to get the best phenacetin results
Phenacetin synthesis method 1
It is obtained by acetylation of p-aminophenethyl ether. The mixture of benzene, acetic anhydride and p-aminophenethyl ether was heated in oil bath for 4 h. After the reaction, the reactant was cooled, that is, finatine was precipitated, filtered, washed with cold benzene and dried. The yield was 86% of the theoretical amount.

Phenacetin synthesis method 2
The product is obtained by the reaction of acetaminophenol and sodium ethoxide. First, acetaminophenol is added to sodium ethoxide, then Iodoethane is added slowly, heated and refluxed for 1h, cooled and filtered. The crude product is dissolved in ethanol, filtered, and the filtrate is diluted with hot water, then cooled, filtered and dried. The yield is 80% of the theoretical quantity. In the first method, acetic acid can be used instead of benzene. The process is as follows. Heat 40% dilute acetic acid to boiling point, add p-aminophenethyl ether, evaporate water, add glacial acetic acid when the temperature rises to 150 ℃, reflux for 1 h, and continue steaming. When the temperature rises to above 150 ℃, take samples to measure free aminophenethyl ether, add acetic anhydride according to the amount of residual aminophenethyl ether, reflux for 0.5 h, check the end point, reduce pressure after passing, and recover acetic acid until the amino content is below 0.046 and the acid content is below 0.2%, The reaction was hydraulically put into the hot refining mother liquor, stirred and cooled to 40 ℃, and filtered to obtain the crude phenacetin. The crude product of phenacetin is dissolved by boiling water or refined mother liquor, heated and stirred, filtered, the filtrate is adjusted to pH = 4.5-4.7 with acid, activated carbon and sodium thiosulfate are added, decolorized, filtered, the filtrate is cooled and crystallized, and then filtered and air dried to obtain phenacetin.
